Albano,Ana Paula Neuschrank; Klafke,Gabriel Baracy; Brandolt,Tchana Martinez; Da Hora,Vanusa Pousada; Nogueira,Carlos Eduardo Wayne; Xavier,Melissa Orzechowski; Meireles,Mário Carlos Araújo.
Paracoccidioides brasiliensis is the etiological agent of the major systemic mycosis in Brazil, called paracoccidioidomycosis. Although the Rio Grande do Sul is considered an endemic area of the disease, there are few studies on the ecology of P. brasiliensis in the state. Therefore, this study aimed to evaluate the infection of P. brasiliensis in horses from the mesoregion of Southwest Riograndense, using these animals as sentinels. Serological techniques, such as double immunodiffusion in agar gel (AGID) and indirect ELISA, were performed to detect the anti-gp43 P. brasiliensis antibody in horses from five different farms in the region of Bagé, RS, Brazil. Serology was performed in 200 Pure Blood English horses up to two years of age that were born and...

Serologic survey for ophidian paramyxovirus in captive snakes MV&Z
Paiva, Maria Isabel Sousa; Araújo Junior, João Pessoa; Barraviera, Benedito.
The captivity maintenance of snakes is a challenge for the breeders, especially in diseases control. The most common diseases in serpentarium and private collections are the parasites, gastrointestinal problems and respiratory viruses. The biggest concern is the ophidian paramyxovirosis caused by ophidian paramyxovirus (OPMV), which primarily affects the respiratory system and may also involve the nervous system. Often fatal, it was described in Colubridae, Elapidae, Viperidae, Boidae and Pitonidae. This virus is transmitted by aerosols, respiratory secretions, contaminated utensils, ectoparasites such as ticks and digestive excreta. There is no specific treatment or vaccine available. The diagnostic methods are: virus isolation in cell culture; electron...

Neospora caninum and Cryptosporidium parvum are intracellular protozoan parasites that are distributed worldwide and of major economical concern in cattle industry. N. caninum can cause abortion storms and high culling rates, whereas C. parvum has zoonotic implications and can cause diarrhea in calves. There are currently no data on the prevalence of neosporosis and cryptosporidiosis in humans or animals in southern Egypt. Prevalence of these two infections was determined in a sample of cattle from two different areas in southern Egypt, Sohag and Qena, using enzyme-linked immunosorbent assay. A total 301 cattle were sampled, of which 18.9% were positive for N. caninum, 35.9% were positive for C parvum and 10.0% were positive for both. Geographical location...

Barbosa da Silva,Jenevaldo; Baêta,Bruna de A.; Lopes,Cinthia T.A.; Martins Land Manier,Bruna Sampaio; Nunes Santana de Castro,Gustavo; Nunes dos Santos,Priscilla; da Fonseca,Adivaldo Henrique; Diomedes Barbosa,José.
Sera samples were collected from 330 water buffaloes on Marajó Island, state of Pará, Brazil, to assess the presence of antibodies against Borrelia burgdorferi by indirect Enzyme Linked Immunosorbent Assay. Approximately 45% of the animals had antibodies against B. burgdoferi. The prevalence of seropositive buffaloes, 72% (85/118), was statistically higher in the city of Soure than in the other municipalities tested. Murrah breed animals were significantly more seropositive (Prevalence Ratio (PR) = 1.84, p = 0.000) than those of the Mediterranean breed. Among the animals diagnosed positive for tuberculosis, 33% (4/12), were also seropositive for B. burgdoferi. Animals positive for tuberculosis had a significantly lower level of B. burgdorferi...

Serological responses in sheep injected with plasmids encoding bovine herpesvirus 1 (BHV-1) gD glycoprotein Arq. Bras. Med. Vet. Zootec.
Cândido,A.L.; Resende,M.; Bessa,L.R.G.; Leite,R.C..
A genetic vaccine consisting of the bovine herpesvirus-1.2a (BHV-1.2a) glycoprotein D (gD) gene under the control of the cytomegalovirus immediate-early promoter/enhancer was generated and administered to sheep intramuscularly in the neck. All animals developed serum antibodies which recognized the homologous antigen (BHV-1.2a strain BH-83) and also exhibited cross-reactivity against the heterologous antigen (BHV-5 strain EVI-190). Three intramuscularly injections were given but serological responses were not improved after the second inoculation. Specific antibodies were detected against BHV-1.2a until at least 12 months after the first inoculation. However, the capacity to induce antibodies against BHV-5 was lower and of shorter duration than to BHV-1.2a.

SEROLOGICAL SCREENING FOR INFECTIOUS CATTLE DISEASES: II. ASSOCIATION BETWEEN PREVALENCE AND LEVEL OF ELISA RESPONSE Ciência Rural
Rojas,José Alfonso Barajas-; Riemann,Hans; Franti,Charles.
A herd of cattle (Holstein-Zebu crosses) was screened every two months by ELISA during a period of two years for IgG antibodies against 19 infectious disease agents. Two hundred and ninety five sera were collected from 157 young animals (0-4 months of age), 1037 sera from 292 developing animals (4-36 months of age) and 1468 sera from 259 producing animals (> 36 months of age). The results indicate that the difference in ELISA between positive and negative tests is associated with the overall prevalence of positive tests. When the prevalence of positive tests is low the difference between positive and negative tests is greater than when the prevalence is intermediate or high. This means that ELISA, presumably other serological tests for IgG antibodies,...

SEROLOGICAL SCREENING FOR INFECTOUS CATTLE DISEASES: III. CHOICE OF SENTINEL ANIMALS Ciência Rural
Rojas,José Alfonso Barajas-; Riemann,Hans; Franti,Charles.
A herd of Cattle (Holstein-Zebu crosses) was screened bimonthly for two years by ELISA for IgG antibodies to infectious disease agents. The herd was composed of 3 age groups: Young animals ( < 4 months of age), developing animals (4-36 months of age) and producing animals ( > 36 months of age). Ali young animals received colostrum within two hours after birth. The overall consistency of ELISA results, i. e agreement between two consecutive tests was about the same for the three age groups and was lowest for infections of intermediate prevalence (40-50%). The interpretation of changes in seroprevalence is most straight forward for young animals. Young animals positive at their first test reflect the immune status of their mothers while young...

Meireles,Luciana Regina; Galisteo Jr.,Andrés Jimenez; Andrade Jr.,Heitor Franco de.
Toxoplasmosis is one of the most prevalent parasitic infections of man and livestock, and its transmission has usually been attributed to ingestion of undercooked or raw meat from infected livestock, with the infection rate in those animals being an important risk predictor of human disease, high in Brazil and São Paulo State. Looking for this public health problem, we tested serum samples from cattle, goat, sheep and chicken from the State of São Paulo, Brazil, for IgG antibodies to Toxoplasma gondii by enzyme-linked immunosorbent assay (ELISA). Antibodies to Toxoplasma gondii were found in 31.00% (62/200) of sheep, 17.00% (34/200) of goat and 11.00% (22/200) of cattle, without positive sample in chicken. Despite differences in feeding habits of each...

Serum kinetics of Calloselasma rhodostoma (Malayan pit viper) venom components in a rabbit J. Venom. Anim. Toxins incl. Trop. Dis.
Tan,NH; Ponnudurai,G; Fung,SY.
The serum kinetics of Calloselasma rhodostoma (Malayan pit viper) venom - specifically two of its components, the major hemorrhagin (rhodostoxin) and a thrombin-like enzyme - was examined in a rabbit by double-sandwich enzyme-linked immunosorbent assay (ELISA). The animal received intramuscularly a 1.0-mg/kg dose of C. rhodostoma venom. The venom level in serum peaked 12 hours after the injection, followed by a gradual decline and finally reached low rates 72 hours after administration. The serum kinetic profile of venom components, however, did not correspond to the profile of the whole C. rhodostoma venom. The serum levels of the C. rhodostoma thrombin-like enzyme increased slowly and peaked only 48 hours post-injection. Then both thrombin-like enzyme...

Serum regucalcin is a useful indicator of liver injury severity in patients with hepatitis B virus-related liver diseases BJMBR
Wei,Xinhuan; Yu,Haibin; Zhao,Peng; Xie,Li; Li,Li; Zhang,Jing.
Regucalcin is a soluble protein that is principally expressed in hepatocytes. Studies of regucalcin have mainly been conducted in animals due to a lack of commercially available kits. We aimed to develop an enzyme-linked immunosorbent assay (ELISA) to quantify serum regucalcin in patients with hepatitis B virus (HBV)-related disease. High-titer monoclonal antibodies and a polyclonal antibody to regucalcin were produced, a double-antibody sandwich ELISA method was established, and serum regucalcin was determined in 47 chronic hepatitis B (CHB) patients, 91 HBV-related acute-on-chronic liver failure (HBV-ACLF) patients, and 33 healthy controls. The ELISA demonstrated an appropriate linear range, and high levels of reproducibility, sensitivity, specificity,...

Stabilising selection on immune response in male black grouse Lyrurus tetrix ArchiMer
Soulsbury, Carl D.; Siitari, Heli; Lebigre, Christophe.
Illnesses caused by a variety of micro- and macro- organisms can negatively affect individuals' fitness, leading to the expectation that immunity is under positive selection. However, immune responses are costly and individuals must trade-off their immune response with other fitness components (e.g. survival or reproductive success) meaning that individuals with intermediate response may have the greatest overall fitness. Such a process might be particularly acute in species with strong sexual selection because the condition-dependence of male secondary sexual-traits might lead to striking phenotypic differences amongst males of different immune response levels. We tested whether there is selection on immune response by survival and reproduction in...
